Abstract

The objectives of this proposal are to exploit two recent major technical breakthroughs, efficient in vivo transposition and rapid PCR-directed gene disruption, to begin to construct a comprehensive, defined knockout library for all non- essential genes in S. coelicolor, and to apply the new technologies to (i) explore the secondary metabolic potential of the genome (ii) analyse gene expression and metabolism under oxygen limitation (iii) define all the genes required for morphological differentiation, and (iv) characterise the Tat secretion pathway. (Joint with grants 88/EGH16081 and 208/EGH16080).
