Abstract

To date, attempts to repair or regenerate cartilage have met with limited success at considerable socio-economic cost. Nonetheless, the avascular nature of cartilage and the fact that it comprises just one cell type makes it an attractive target in the area of tissue engineering. We propose that by using selectively isolated and expanded human mesenchymal stem cells in combination with novel peptide-based self- assembling scaffolds within purpose-built bioreactors, it will be possible to create constructs ideally suited to cartilage formation. To achieve this, we describe a unique multidisciplinary research programme across three centres that harnesses expertise in: i) mesenchymal stem cell technology (Southampton); ii) peptide design and self- assembly and nanobiotechnology (Sussex); and iii) bioreactor design, development and tissue modelling (Bath).
