Abstract

We have solved the structure of the explosive degrading enzyme PETN reductase at high resolution and also specific complexes with inhibitors and explosive molecules. We will now exploit this structural information by investigating the novel mechanisms of explosive degradation using a combination of site-directed mutagenesis, X-ray crystallography, transient kinetic and solution studies. We will probe the role of Trp-102, which forms a novel covalent link with nitroaromatic explosives, and several other residues implicated in catalysis. We will also address the chemical mechanism of triazine explosive degradation and investigate the multiple chemical mechanisms available from nitroaromatic degradation. (Joint with grant 8/C13574.)
