Abstract

A variety of techniques will be employed to provide suitable crystals of hepatitis B core particles to obtain a high resolution structure. This will then be used to direct mutagenesis experiments designed to investigate the factors influencing core particle assembly, virus RNA/pol encapsidation and the surface protein associations which promote virion assembly. An understanding of these interactions at the atomic level should be valuable for the design of potential antiviral inhibitors. Also, chimeric cores including genetically fused epitopic domains from foreign sources will be examined to understand details of antigenic structure and to assist in the design of novel recombinant vaccines. Finally, core particles decorated with receptor binding domains from other viruses and proteins will be used to study ligand/receptor interactions and to identify inhibitory molecules.
