Abstract
The progressive accumulation of senescent mammalian cells is a plausible biological mechanism for the ageing of regenerative tissue. Understanding the mechanisms by which cells enter senescence will provide valuable insights into the means by which they accumulate in ageing bodies. Strong evidence exists that senescence in some cell types is telomere dependant. However, there is also good evidence from both humans and rodents that a telomere-independent pathway exists. The applicants seek support to determine the relevance of both pathways in model human cell systems and to evaluate methodologies aimed at achieving the stable ablation of senescence.
