Abstract
The aim of the proposed work is to use a combination of in vitro and in vivo models to test the hypothesis that senescent cells contribute to aged pathology. Acellular fibrous dermal collagen (FDC) preparations maintain typical matrix architecture. By culturing young of senescent rat dermal fibroblasts with FDC derived from young or geriatric rats, or FDC seeded with young or senescent cells, will be grafted into full thickness loss skin wounds in chests of young rats. Histological and biochemical analyses of the grafts after two and four weeks in situ will reveal whether cell age or matrix age is the main cause underlying the changes that are characteristic of aged skin.
