Abstract

Gene expression in Lawsonia intracellularis will be examined during infection in vitro in intestinal cell lines and in vivo by use of a promoter probe plasmid system. An adaptation of IVET/DFI methods with CAT and GFP as dual reporters in a broad host range plasmid will be applied to this highly fastidious obligate intracellular pathogen. Chloramphenicol and/or ampicillin will be used to select and assess gene expression. The system will be employed to monitor expression of an identified Lawsonia gene (a haemolysin which we have characterised) as well as global examination of expression. These investigations will characterise loci which are essential and/or up-regulated during infection. The defining of these determinants has multiple downstream applications.
