Abstract

The project combines the complementary expertise of two laboratories (Manchester and Sheffield) in an investigation of the role of Parathyroid Hormone related Protein (PTHrP) as a hypercalcaemic factor in teleost fish Ca homeostasis. Using the euryhaline flounder in vivo studies will establish the tissue source of circulating PTHrP and physiological control of PTHrP secretion, blood peptide concentration and target tissue action. Isolation and sequencing of PTHrP and homologous RIA development will be coupled with cloning of flounder genes for PTHrP, PTHrP receptor and the calcium-sensing receptor (CaR). Tissue PTHrP and PTHrP-receptor expression will be established along with possible coincident CaR expression. The CaR mediation of the modulatory role of blood [Ca] on both PTHrP secretion and PTHrP target tissue action will be defined by in vivo and in vitro studies. (Joint with 50/S09874).
