Abstract
We have identified a signal transduction system that modulates expression of cell envelope-related genes in response to cell envelope stress in S. coelicolor. The system is comprised of four proteins; sigmaE, an RNA polymerase sigma factor; CseB, a response regulator; CseC, a sensor kinase; and CseA, a negative regulator of unknown biochemical function. The aims of the proposed work are to define (i) the biochemical function and (ii) the subcellular location of CseA (iii) to analyse the eps operon under sigmaE control (iv) to characterise the product specified by eps operon (v) to define the CseB binding site(s) on the sigE promoter (vi) to identify further genes under sigmaE control, and (vii) to improve the commercial potential of the sigE system as a basis for a broad-range, generic screen for cell envelope-specific antibiotics.
