Abstract

Normal seed development in flowering plants requires a ratio of 2 maternal to 1 paternal genomes in the endosperm, reflecting the existence of parental imprinting. We have shown that in Arabidopsis, altering the parental genome balance disrupts endosperm development (Scott et al. 1998). In particular, several aspects of the cell cycle - rate of mitosis, duration of proliferation, and timing of cytokinesis - are dramatically affected in endosperm with extra maternal or paternal genomes. We propose to study the interaction between cell cycle, seed development, and imprinting. Expression of cell cycle genes in developing seeds will be investigated, and imprinted genes will be identified using allele-specific RT- PCR and in situ hybridisation. (Joint with award P12018.)
