Abstract

This project is aimed at investigating the anaerobic biosynthesis of vitamin B12 in Bacillus megaterium and Salmonella typhimurium. Using recombinant Escherichia coli strains harbouring expression plasmids containing the cobinamide biosynthetic genes from these organisms, a structural analysis of accumulated intermediates will be undertaken, thus elucidating the step-by-step conversion of precorrin-2 into cobinamide. This will clarify (i) the point at which cobalt enters the path, (ii) the method employed to cause ring contraction and (iii) the role of some of the proteins in these operons with no ascribed function.
