Abstract

(1) To determine the 3-dimensional structure of enoyl reductase (ER) from rape using X-ray crystallography and to identify residues important in substrate binding and catalysis. (2) To carry out a detailed kinetic analysis of ER. (3) To identify important residues in ER through a programme of covalent modification and site directed mutagenesis. (4) To investigate the molecular basis of the reaction between ER and its acyl carrier protein substrate. (5) To produce large quantities of B-ketoreductase (B-KR) from rape in order to refine the conditions for crystallisation and for crystallisation trials and kinetic analysis.
