Abstract

In previous work we have developed tools and methodology which allow genomic mapping in chickens, with the long term aim of mapping genes involved in disease resistance. We have now used these tools to map novel genes for resistance to salmonellosis and to Mareks disease. These are the first genes to be mapped in this manner in chickens and are of considerable commercial and scientific importance. The successful mapping of these genes strongly suggests our genomic mapping approach can be applied to other disease resistance and immunological traits, and we have already identified genetic differences in these traits in preparation for this work. However our existing 373A automated sequencer is already in use continuously. The purchase of an ABI 377 sequencer will greatly expand our capacity for automated genotyping of microsatellite and AFLP loci and will allow us to analyse further disease resistance genes and also the larger numbers of animals necessary to investigate genetically more complex traits. Greater sequencing capacity will also allow us to analyse allelic differences in genes already identified, making it possible to develop efficient selection methods for variants in these genes and to greatly extend our present work relating the molecular effects of sequence differences to phenotype.
