Abstract

Funds are sought for a Beckman XL-I analytical ultracentrifuge. The instrument will be used for a variety of biological research projects. For example, a number of researchers at Sussex are interested in roles played by coiled-coil motifs in directing and cementing protein-protein interactions. Coiled coils from dimers, trimers and some higher-order structures, and the principal solution method for determining with oligomer states are present is sedimentation-equilibrium analysis performed in the analytical ultracentrifuge. The group includes Dr Woolfson, who has considerable experience in the prediction, design and characterisation of coiled coils. Projects in this area include: the characterisation of non-canonical coiled-coil motifs; the design of new coiled coils; the structural analysis of vesicular- fusion proteins (SNAREs); the characterisation of novel bZIP transcription factors involved in neuronal regeneration; the molecular dissection of large coiled-coil-containing. DNA-repair proteins. The provision of an analytical ultracentrifuge at Sussex would benefit other biochemists who are researching protein folding, protein design and various macromolecular interactions, and chemists with interests in polymer science. An instrument would be housed in a purpose-built room and maintained by a technician experienced with ultracentrifuge.
