Abstract

This equipment grant application is a request for funds to set up a multifunctional blot and gel imaging system that will provide phosphorimaging, fluorescence and quantitative densitometry facilities in a single package. Three separate projects are described in detail which will make use of these facilities. Each of these will greatly benefit from the increased speed and sensitivity, large dynamic range and accurate quantitative nature of the imaging capabilities of the system. These are: 1) Identification of the physiological eIF4E kinase from Xenopus laevis eggs and serum- stimulated cells; Quantification of the level of phosphorylation of eIF4E in response to cell growth; Mapping of the mitogen-stimulated phosphorylation sites on eIF4G. 2) Cell cycle control in B lymphocytes. 3) Studies on membrane protein assembly.
