Abstract

The current goal of the IAH is to understand host-pathogen interactions at a molecular level. Many important proteins have been sequenced, purified and used to generate antibodies. Understanding how and where these proteins function in the host requires the sub-cellular localisation of both host and pathogen proteins. To undertake this work, there is an urgent need for improvements of the Institute's electron microscopes. We request one TEM to replace two current TEMs. One is 25 years old and the other is 15 years old and inappropriate for our present and future needs. The new microscope will be sited at Pirbright in the Bioimaging unit which has been recently established. Experienced staff have been recruited and the unit is fully equipped for specimen preparation. The unit will underpin the science of the Institute of all three sites. This application details nine projects where cryoTEM or immunolabeling will be essential to their progress and covers a protozoan, bacteria and several virus projects. In each case, virulence factors have been characterised at a molecular level. Understanding the cell biology of how and where they act in the host cell requires high-resolution localisation by electron microscopy. This application justifies the purchase of a new TEM.
