Abstract
In this study we will develop a new range of non-viral vectors for gene transfer, based on branched dendrimer molecules. We will investigate the potential for targeting dendrimers using ligands, peptides and antibodies, and also to evaluate two new types of molecule (dendron and unsymmetric dendrimers), optimising features of the design of these molecules such as their geometry, flexibility and internal free space. The project offers the possibility of new generic technologies for gene transfer, including targeted gene delivery. These will be of benefit not only in the context of gene therapy, but also for experimental testing of novel gene constructs prior to developing transgenic animals, for example for xenotransplantation.
