Abstract

There is a far greater than expected developmental plasticity in mammalian cells, including that shown after nuclear transfer. However, in present procedures only a small proportion of cells or nuclei are reprogrammed appropriately. In this collaborative application we propose to investigate the ability of oocyte and egg extracts to reprogramme mammalian nuclei before either cell culture or nuclear transfer. We will overcome the limitation imposed by the small quantity of material available from mammalian eggs by taking advantage of the ability of frog extracts to reprogramme gene expression in mammalian nuclei. Reprogramming will be assessed by observations of gene expression and of the ability of treated cells to support development after nuclear transfer. (Joint with grant number 20291).
