Abstract

A growing body of evidence exists to suggest that reactive oxygen species (ROS) impact on the life span of multicellular organisms, by causing the accumulation of damaged macromolecules in post-mitotic cells. The aim of this project is to test the hypothesis that the capacity to synthesise the endogenous antioxidant glutathione has a major influence on life span by protecting against the deleterious effects of ROS. We propose to manipulate the activity of the rate-limiting enzyme in glutathione biosynthesis, gamma- glutamylcysteine synthetase, in the fruit fly Drosophila to create animals with both increased and reduced capacity to synthesise glutathione. The effects on resistance to oxidative stress and life span will be examined. (Joint with grant 108/G15090).
