Abstract
We propose a unique multi-disciplinary approach to improve significantly the current technology for gene delivery to stem cells and cells using a non-viral delivery system with a macroscopic controlled release scaffold. The use of macroscopic controlled released scaffolds will provide sustained gene delivery and, critically, functionality over the entire period of tissue regeneration/repair within defined nanoscale architecture. Gene delivery will be achieved using cationic polymers as non-viral vectors. This combination is achievable through the use of supercritical fluid mixing technology to generate, ultimately, scaffolds encapsulating the gene vehicle and, significantly, functionality of the gene delivery system. The generation of functional bone from this proposal has significant healthcare implications and the generation of non-viral gene delivery and scaffold release technology will have widespread tissue engineering applications (Joint with grant 18597).
