Abstract

The aim is to generate high affinity inhibitors by using a protein as a template to assemble dimeric ligands in situ. This approach will facilitate the delivery of high molecular weight multimeric ligands to their site of action. Two strategies will be pursued: connection of the ligands via metal chelate coordination and via disulphide bond formation. We propose to use calmodulin as a model system for the development of general methodology for the delivery of high molecular weight multivalent ligands which self-assemble at their site of action and bind to multiple sites on a biological receptor.
