Abstract

The chemical synthesis and biotransformation of alkylaryl and dialkyl 1,2-disulfides to yield enantiopure thiosulphinates using dioxygenases (and monooxygenases) will be studied. This asymmetric sulphoxidation study will be complemented by a kinetic resolution approach using racemic thiosuphinate and sulphinate substrates in the presence of sulphoxide reductase enzymes. The synthetic potential of these thiosuphinate and sulphinate enantiomers, will be evaluated with particular emphasis on nucleophilic displacement reactions. The formation of transient thiophene oxides as metabolites, and their potential as in vivo dienophiles to trap diene intermediates (cis-dihydrodiols, triols) will be studied. A comparative study of the formation of thiophene oxides using chemical and enzyme (monooxygenases and dioxygenases) methods, and their synthetic value in the context of polycyclic thiaarene chemistry is proposed.
