Abstract

This work will develop and exploit methods of controlling and measuring the ionisation state of enzymes in low water media. In particular we will explore the application of newly discovered solid-state acid-base buffering systems in synthetically useful organic phase biotransformations. Potentially these solid buffers can eliminate detrimental changes in enzyme activity or enantioselectivity arising from reactants or products with acid-base properties. This will be tested on reactions leading to biologically active pharmaceutical intermediates such as the dynamic resolution of 5(4H)-oxazolones by lipases and deprotection of phenylacetamides by penicillin amidase. Close collaboration between Strathclyde and Edinburgh will ensure that new insights into the physiochemical control of enzyme catalysed reactions will be rapidly adopted in synthetic applications.
