Abstract

Epidemiological and physiological studies have provided substantial evidence that consumption of crucifers significantly reduces the risk of cancer. Several possible mechanisms have been proposed, many of which implicate glucosinolates and their degradation products, which include isothiocyanates (ITCs) and indolyl compounds. We propose to study changes in gene expression in human colon cell cultures with Affymetrix Human GeneChipTM U133 microarrays, following treatment with extracts of broccoli and isothiocyanate-enriched broccoli, and pure compounds. We will also compare the effect of broccoli extracts on gene expression with that of Arabidopsis extracts, and evaluate the potential to develop Arabidopsis as a model system to study the anticarcinogenic effect of crucifers due to its amenability to genetic modification.
