Abstract

Cells discriminate between metals to ensure that the correct ions locate to the correct proteins and prosthetic groups but, whilst the mechanisms responsible are fundamental to cellular viability, the actual processes mostly await discovery. By using Synechocystis PCC 6803 as a model system for a study of the relationship between metal requirement and tetrapyrrole biosynthesis, we plan to investigate the in vivo and in vitro activity of the ferrocheletases and cobaltochelatase found in this organism and address the problem of how these enzymes discriminate between divalent metal cations and acquire their metal substrate. In the cases of cobalt we will also study the relationship with metal export and aim to identify tetrapyrrole-derived signalling molecules that help to control and regulate metal homeostasis.
