Abstract

RAMPs form heterodimers with the G-protein-like receptor CRLR to yield receptors for CGRP and adrenomedullin. We will analyse the structure of RAMP1 by site directed mutagenesis and production of chimeric receptors with RAMP3 to identify key regions needed for glycosylation, structural integrity and the ability to interact with CRLR to produce a functional CGRP receptor. We will also investigate the conditions needed to produce a correctly-folded, soluble version of the extracellular domain of RAMP1 and characterise this with respect to both its structure (NMR, CD, FT-IR) and functions (interaction of CRLR to produce a CGRP receptor.
