Abstract

This proposal focuses on ABC50, an ATP-binding cassette protein which we recently discovered to interact with translation initiation factor eIF2, which is important in controlling protein synthesis. ABC50 stimulates the binding of Met-tRNA to eIF2 and also binds to ribosomes. ABC50 is thus a novel component of the mammalian translational machinery, and this proposal aims to clarify its function in mRNA translation. We will (i) use a range of approaches to explore the role of ABC50 in translation, in particular in events that involve eIF2; (ii) analyse how ABC50 stimulates eIF2 activity; (iii) identify the region of ABC50 that binds eIF2 and (iv) study the role of nucleotide binding/hydrolysis in the function/interactions of ABC50.
