Abstract
Control of smooth muscle contraction and many forms of cellular motility relies on regulation of the motor protein myosin, yet the structural mechanism of this control is unknown. Using low dose electron microscopy, including frozen hydrated unstained specimens, large numbers of high resolution images will be recorded of regulated myosin and modified recombinant molecules in the on and off states. Computer analysis by single particle averaging will reveal consistent details in the structure while suppressing random variations. Comparison of on and off states with each other and the atomic structure of the isolated head will reveal the differences within the myosin heads and in their arrangement that underlie the switching mechanism.
