Abstract

To advance the design and operation of selective adsorptive recovery of retroviral vectors from animal cell cultures, via: (1) The selection of immuno-chemical reagents that selectively interact with virions with association/dissociation constants suited to the adsorptive recovery of vectors from complex feedstocks. (2) The identification of solid phases and associated contactors having sufficient physical capacities and viral accessibility to efficiently accommodate viral particles adsorbed from complex feedstocks. (3) The controlled covalent immobilisation of reagents from (1) to the surface of solid phases from (2) to achieve efficient adsorption and desorption of retroviral agents. (4) The testing of adsorbent assemblies in the recovery of retroviral vectors from realistic production feedstocks in comparison with more conventional adsorptive (e.g. ion exchange and hydrophobic chromatography) and size- based (e.g. density-gradient centrifugation and gel permeation) technologies.
