Abstract

Actinomycetes, especially Streptomyces species, are major producers of commercially important secondary metabolites. A strain of S. coelicolor has been developed for in vivo cosmid packaging into phiC31 phage particles. This project aims to optimise cosmid packaging in vivo and to develop conditions for in vitro packaging. In addition, the host range of cosmid delivery will be investigated and the feasibility of expression in antibiotic biosynthesis genes or global regulators in many different species will be demonstrated. These new genetic tools should facilitate screening of combinatorial, directed evolution or metagenome libraries in many different genetic backgrounds for synthesis of novel secondary metabolites.
