Abstract
This five year programme is proposed as a continuation of our present MRC programme grant. The scientific aims are to define key events in replication and transport of the infectious agent and the role of PrP in host susceptibility. The information derived from this programme will provide a basis for developing strategies for therapeutic intervention into these diseases. We aim to address the following fundamental questions: How do mutations and polymorphisms in the human PRNP gene alter disease susceptibility? What is the role of the follicular dendritic cells in agent replication? How does TSE infectivity spread to and within the CNS? Does PrP glycosylation determine strains and strain targeting?
