Abstract

To use gene targeting in embryonic stem cells to produce new mouse strains with specific alterations to their endogenous prion protein (PrP) genes. Mice with mutations found in Gerstmann-Straussler- Scheinker Syndrome and Creutzfeldt- Jacob Disease will be studied for the development of spontaneous neurodegenerative disease and for susceptibility to scrapie. Mice with PrP codon 108 and 189 changes will be used to resolve the uncertainty over the equivalence of the PrP and scrapie incubation time (Sinc) loci. Mice with truncated forms of PrP will be used to investigate the relationship between PrP protein structure and ability to support replication of the scrapie agent.
