Abstract

The aim is to exploit our newly developed methodology for photocaging proteins to examine protein folding on ultrafast timescales. We will use our cross-linking reagents to constrain proteins in non-native conformations and initiate folding reactions using laser flash photolysis. A unique feature of this approach is that we will be able to vary the starting conformation in a controlled manner, characterise its structure with very high positional resolution using heteronuclear NMR methods, and synchronise all molecules in this state. We will be in a position to answer very specific questions about the nature of the free energy landscape that controls protein folding reactions, down to picosecond timescales.
