Abstract

We have discovered that in the early phase of adenovirus (Ad) infection of human cells, E1A proteins co-localise with Cajal bodies (CBs). We will map the domain of E1A that mediates the interaction with CBs using a panel of recombinant Ads containing small, in-frame deletions in E1A. The CB protein interacting with E1A will be defined by reversible protein cross-linking using CBs purified from Ad-infected cells followed by deletion mutagenesis. The importance of the E1A: Cajal interaction for establishment of productive viral infection will be investigated by targeted disruption techniques. We will analyse the dynamic association between E1A and the CB using time-lapse microscopy techniques. Finally we will also investigate whether transcriptional regulatory proteins of other DNA tumour viruses (namely HPV and SV40) interact with the Cajal body.
