Abstract
1) Identification of functional origins of replication in the archaeon M. thermoautotrophicum. 2) Inclusion of origin sequences in vectors for use in the development of a transformation protocol for this organism. 3) Production of an in vitro DNA replication system based on archaeal components. 4) Elucidation of archaeal initiation, as a paradigm for eukaryotic replication.
