Abstract
The objectives of this proposed research programme are: 1) identification of functional origins of replication in the archaeon M. thermoautotrophicum; 2) inclusion of origin sequences in vectors for use in the development of a transformation protocol for this organism; 3) production of an in vitro DNA replication system based on archaeal components; and 4) elucidation of archaeal replication initiation, as a paradigm for eukaryotic replication.
