Abstract

Tubulin is an important target for cancer chemotherapy. Drugs that bind to the protein can block the dynamic equilibrium that exists between tubulin dimers and microtubules, resulting in cell death. Colchicine is an antimitotic agent known to inhibit tubulin polymerisation. The project will try to understand the detailed interactions involved between tubulin and bioactive ligands that bind to the colchicine binding site. The study of this protein:ligand interaction should allow the development of a molecular model for the colchicine-binding site followed by the design and biological evaluation of novel agents capable of binding specifically to tubulin with reduced side effects.
