Abstract

Transcriptional activation is a fundamental process controlling gene expression. A large family of enhancer-dependent transcriptional activators including NtrC and PspF belongs to the AAA plus (ATPase Associated with various cellular Activities) super family of proteins. AAA plus proteins are required for numerous cellular activities in all life forms. The ATPase activity of the AAA plus transcription activators have been shown to target a key basal transcription complex and remodel it. Here we propose to characterise the nucleotide- dependent oligomerisation states of the AAA plus transcription activators, e.g. NtrC and PspF, as well as the associated holoenzyme using X-ray crystallography and cryo electron microscopy. This study would not only form foundations to understand the molecular mechanisms of the AAA plus transcriptional activators but also add essential structural information to the diverse AAA plus family.
