Abstract

Molybdopterin (MPT), the ligand of Mo or W at the catalytic centre of enzymes, will be synthesised and characterised. Dithiolene chemistry will be developed: analogues of the enzymes' catalytic centres will be produced for MPT and relatives; the physical and chemical properties of these systems will be determined. Binding of the synthetic MPT, including nuclear spin labels, to Cnx1 (a key protein in the biosynthesis of Moco in plants) and the incorporation of synthetic MPT into an enzyme will be studied. Binding of Mo and W by Cnx1 and insertion into MPT will be monitored. Studies with R. sphaeroides will produce W-substituted enzymes and determine how: i) MPT acquires a nucleotide and ii) two molecules of MPT are bound to the metal. (Joint with grant 34/B15270).
