Abstract

The discovery of new improved antibiotics remains a major global concern. Recently we have identified and cloned a region of the Streptomyces coelicolor genome that encodes the enzymes required for the production of the Calcium-Dependent Antibiotic CDA, a complex cyclic lipopeptide. By a combined biochemical and chemical approach we will: 1) Elucidate the biosynthetic origins of CDA; 2) Determine the function of the non-ribosomal peptide synthetases and other enzymes required for CDA biosynthesis; 3) Establish a set of rules for engineering the biosynthesis of novel lipopeptides in the model actinomycete host.
