Abstract

This collaborative study, based on UB-165, a novel, enantiospecific subtype- selective nicotinic agonist, aims to define individual pharmacophores for neuronal nicotinic receptor (nAChR) subtypes, and to use the ligands to characterise native nAChR. A series of structural analogues of UB-165, as well as new classes of ligands, will be generated by efficient chemical synthesis. All ligands will be characterised pharmacologically at defined nAChR subtypes, and used to distinguish presynaptic subtypes in the brain. These results will direct ligand design, generating informative structure-activity relationships and the evolution of new, more critical pharmacophore models that will be significant for rational drug design targeted at these sites.
