Abstract

We propose to develop a strategy for the conformational analysis of oligosaccharides and oligosaccharide protein complexes by use of oligosaccharide ligands >95% enriched with 13C in combination with multidimensional NMR techniques at low sample temperature. The overall aims of the proposed project are as follows: (1) Application of novel multidimensional, multinuclear NMR techniques at low temperature for characterisation of structure and dynamics of 13C-enriched oligosaccharides in solution. (2) Incorporation and optimisation of these techniques in TRNOE studies on a model carbohydrate-lectin complex. (3) Application of the optimised techniques to a carbohydrate-protein interaction of topical biological interest.
