Abstract
The objectives of this proposed research programme are: (1) to isolate and clone proteins which interact with the c-myc internal ribosome entry segment (IRES). (2) to identify protein partners of the c-myc IRES binding proteins. (3) to investigate expression and regulation of the IRES binding proteins. (4) to determine RNA/Protein interactions between c- myc IRS binding proteins and a range of IRESes. (5) to compare and contrast the c-myc IRES with IRESes in Myc family of genes. Targets to be met are: In 1-18 months: purify and clone c- myc IRES binding proteins. Over-express proteins in bacteria and purify to homogeneity. Establish collaborations with groups who are working on other IRES binding proteins. In 18- 36 months: Identify protein partners and investigate regulation of IRES binding proteins. In 36-48 months: determine RNA protein interactions. In 48-60 months: Studies on other Myc family IRESes.
