Abstract
The aim of this project is to develop efficient and high yielding biocatalysts for the hydroxylation of a range of organic substrates in a predictive manner. These catalysts will be based on P450 enzymes present in Actinomycetes. The strategy to be employed involves high-level expression of genes, evaluation and selection of cloning host, determination of the selectivity exhibited by expressed cell free hydroxylases, development of whole- cell systems that are capable of carrying out efficient hydroxylation reactions, development of protocols for fermentation of organisms and for carrying out whole cell biotransformations based on scale-down studies and the application of these biocatalysts to organic substrates of current interest.
