Abstract

We wish to demonstrate the evolutionary relationship between the plant sphingolipid desaturases and the front-end fatty acid desaturases, the latter being important for the synthesis of nutraceutical compounds such as GLA and arachidonic acid. This will be achieved by mutageneisis of the sphingolipid desaturase, to convert it to a D6 fatty acid desaturase. Functional analysis will be carried out by expression in yeast and Arabidopsis, followed by GC and GCMS of lipid fractions. We will also generate novel fatty acid desaturases by using DNA shuffling of cytoplasmic domains of the sphingolipid desaturase. Identification of new fatty acid desaturase activities generated by shuffling will be selected by a yeast complementation assay.
