Abstract

We propose a wide-ranging and fundamental study of modular polyketide synthases, to establish generic technologies for: (1) rational engineering of known bioactive compounds to make hitherto inaccessible derivatives; and (2) development of an effective biological counterpart of combinatorial chemistry, to generate large numbers of diverse compounds as potential leads in drug discovery.
