Abstract

We propose to study the genetic basis of murine growth by focusing on a major quantitative trait locus (QTL) we have mapped in previous work to a c.1cM interval of the X chromosome. We propose to identify the causal genetic factor(s) underlying the QTL by ultra-resolution mapping, and to identify all of the candidate alleles by sequencing of the minimal genomic region containing the QTL. We shall characterise the physiological effects of allelic variation at the locus by comparing phenotypes of congenic lines that differ only for the QTL region, correlate changes in expression of candidate gene(s) in the minimal QTL region with changes in phenotype, and investigate the overall effect of variation at the QTL on global gene expression.
