Abstract

Phage display antibodies have been developed which recognise GnSAF/IF bioactivity and also clones in a human ovary cDNA library. The single chain antibodies have been converted to human IgGs and will be used to: 1). identify the specific GnSAF/IF protein by immunopurification, Western blot and bioactivity monitoring of 2D gels of granulosa cell-conditioned medium; 2). Characterise the GnSAF/IF protein and gene sequence; and 3). Develop a GnSAF/IF immunoassay using immunopurified or recombinant GnSAF/IF. This project will finally resolve one of the reproductive enigmas of the last 20 years and will provide the basis for the assessment of the importance and potential of GnSAF/IF in both medical and veterinary situations.
