Abstract

We are in a unique position to exploit array technology for investigating genome-wide changes in macrophage gene transcription in response to African swine fever virus infection. This technology will improve our understanding of the pathogenic mechanisms of ASFV. We will construct and use a porcine cDNA microarray, to obtain expression profiles from uninfected macrophages; macrophages infected with ASFV isolates of varying pathogenicity; and macrophages infected with mutants lacking specific genes which interfere with host transcription factors. We will also examine macrophage responses to ASFV in warthogs and bushpigs, which can be infected but show no disease signs. The project will yield vital clues into viral immune evasion strategies, and provide a valuable insight into ASFV pathogenesis and natural host viral suppression mechanisms.
