Abstract

We propose to purify and biochemically characterise the protein kinases secreted by infective (muscle stage) larvae of the parasitic nematode Trichinella spiralis. Genes encoding the secretory protein kinases will be isolated and the primary structure of the enzymes determined. The dynamics of expression and localisation of parasite-derived kinases in infected rat skeletal muscle will be determined. Potential substrates for the enzymes will be investigated in vitro, and the phosphorylation status of proteins which regulate cell cycle re-entry examined throughout infection in vivo.
